Abstract

Agrobacterium tumefaciens strains were identified isolated from the artificial lake water of Sabanci University campus, Istanbul, Turkey. These isolated strains were confirmed as Agrobacterium sp. using various biochemical, pathogenicity (tumor forming ability), and molecular tests in comparing with A. tumefaciens strains C58C1-pmp90. Firstly, 36 bacteria colonies were observed and screened on the basis of their color development on selective media. From these initial results, the isolated bacteria were tentatively identified as Agrobacterium sp. strains. The biochemical features of selected isolates were tested to their ability to utilize several opine types; and from these results, 16 isolated bacteria were identified as Agrobacterium sp. According to pathogenicity test, only four out of isolates including the standard sample, show positive. For 4 isolates including positive control, ti plasmid was isolated and nopaline synthase (nos) region was amplified for 4 isolates, with a fragment of 118 bp. As a result, 2 isolates were determined as Agrobacterium tumefaciens.
